Abstract
A Croton stellatopilosus green callus and a cell suspension culture were established on Murashige and Skoog medium supplemented with 2 mg/L naphthaleneacetic acid (NAA), 0.2 mg/L benzyladenine (BA), and 1 % (w/v) sucrose. The profiling of terpenoid accumulation in the leaf, green callus and cell suspension cultures of Croton stellatopilosus were carried out by using gas chromatography-flame ionization detection (GC-FID). Terpenoids including phytol, geranylgeraniol (GGOH), plaunotol and phytosterols were accumulated in the green callus culture, but phytol and plaunotol were not detected in the cell suspension culture. The transcription profiles for 1-deoxy-D-xylulose-5-phosphate synthase (DXS), 2C-methyl-D-erythritol 4-phosphate synthase (MEPS) and geranylgeranyl diphosphate synthase (GGPPS) were obtained. All three genes were highly expressed in the cells from leaves and of the green callus culture but not in the suspension cells, and this was in agreement with the terpenoid profiling. © 2013 Society for Plant Biochemistry and Biotechnology.
