dc.description.abstractTri-Kesornmas (TK) is a traditional Thai remedy made of coral bush stem bark, lotus stamen, and bael fruit. Based on its traditional use for bringing good health and assistance in healthy aging, this study aimed to investigate the potential in oxidative stress resistance, lifespan extension, and activity against the toxic amyloid-beta (Aβ) protein in Caenorhabditis elegans. The herbal mixture was boiled in water, which was then spray-dried to make a dry powder. It was discovered to include flavonoids and phenolic compounds, with a protocatechuic acid level of 160.83 ± 3.91 μg in 100 mg of TK powder. The effects of survival under oxidative stress conditions and lifespan extension were studied in wild-type N2 worms. When compared to the control, TK at 10 μg ml−1 increased worm survival under oxidative stress conditions (p = 0.0058), and TK at 10 μg ml−1 and 100 μg ml−1 increased lifetime (p = 0.0392 and 0.0020, respectively). Finally, the activity against Aβ toxicity in the transgenic strain CL4176 was assessed by paralysis delay. When TK 100 μg ml−1 was compared to the control, it significantly delayed paralysis (p = 0.0387). It was established that TK lowered oxidative stress, increased lifespan, and decreased Aβ toxicity in C. elegans. © 2022 Roongpetch Keowkase et al.
